Mechanism of Action
Ketoprofen exhibits anti-inflammatory, analgesic and antipyretic properties. It potently inhibits the enzyme cyclooxygenase resulting in prostaglandin synthesis inhibition.
Absorption: Readily absorbed from the GI tract (oral); reduced absorption w/ food. Absolute bioavailability (oral): Approx 90%. Time to peak plasma concentration: Approx 0.5-2 hr.
Distribution: Enters synovial fluid (substantial concentrations). Volume of distribution: Approx 0.1 L/kg. Plasma protein binding: Approx 99% (mainly to albumin).
Metabolism: Hepatic via conjugation w/ glucuronic acid.
Excretion: Mainly via urine (as glucuronide conjugates). Elimination half-life: Approx 1.5-4 hr.
